高温和内毒素联合对RAW264.7细胞凋亡影响

摘要: 目的 研究高温和内毒素复合因素对RAW264.7细胞凋亡的影响。方法 将RAW264.7细胞分成37℃空白(BLANK)组、37℃内毒素(LPS)组、40℃空白组和40℃内毒素组,置于含50%CO2细胞培养箱中,分别于3、24 h后取出,用流式细胞术检测细胞凋亡百分率,并对凋亡细胞生化标志性指标DNA片段进行检测。结果 37℃BLANK3 h组和24 h组细胞凋亡率极低,分别为(3.6±0.7)%和(4.6±1.9)%,同时37℃LPS 3 h组未见明显细胞凋亡现象,凋亡率为(2.8±0.6)%;而40℃LPS组凋亡情况较37℃LPS组严重,其中,40℃LPS 3 h组细胞凋亡率(6.7±0.6)%与37℃LPS 3 h组比较明显增高(P<0.05);40℃BLANK3 h组凋亡率与37℃BLANK3 h组比较虽有所增加,但差异无统计学意义(P>0.05);随着时间的延长,细胞凋亡情况明显加剧,37℃LPS 24 h组、40℃BLANK24 h组和40℃LPS 24 h组细胞凋亡率分别为(13.4±5.5)%、(21.2±2.8)%、(67.4±3.8)%,与同温度不同时间组比较,差异均有统计学意义(P<0.05)。结论 高温和内毒素对RAW264.7细胞具有损伤作用,在高温和内毒素作用下细胞会出现明显凋亡现象,且随着时间的延长,损伤程度逐渐加重,凋亡细胞数量大大增加。Abstract: Objective To study the effect of high temperature combined with endotoxin on the apoptosis of RAW 264.7 cell.Methods The cultured RAW 264.7 cells in vitro were divided into 4 groups:normal temperature(37℃) blank,high temperature(40℃) blank,37℃+lipopoly saccharide(LPS),and 40℃+LPS guoup.After 3 and 24 hours culture under the condition of 95% O2 and 5% CO2,we detected the apoptoisis rate of RAW 264.7 cells with flow cytometry.Results Apoptosis rates were extremely low in 37℃ blank group(both 3hr and 24hr),as well as in 37℃+LPS group.The high temperature(40℃) groups,on the other hand,showed higher possibilities of cell apoptosis.Compared with 37℃+LPS goups,40℃+LPS group showed a much more obvious tendency of cell apoptosis(P < 0.05).But there was no significant difference(P > 0.05) between the apoptosis rate of 40℃ blank group and that of 37℃ group.As time passed by,the rate of cell apoptosis increased markedly (P < 0.05).Conclusion High temperature combined with endotoxin plays an importantrole in the apoptosis of RAW 264.7 cell.And the severity of cell apoptosis rises significantly with the extension of exposure tmie.
