超强静磁场对幼鼠CAT和GSH-Px基因表达影响

Impacts of 12 T ultra strong static magnetic fields on gene expression of catalase and glutathione peroxidase in mice

  • 摘要: 目的 探讨超强静磁场(USMF)对幼龄小鼠过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)基因表达的影响。方法 1周龄小鼠分为3个照射组和对照组,分别经12 T的USMF照射8,12,16 h和假照射处理;照射结束6、12、24 h后提取小鼠肝脏中总RNA,用逆转录-聚合酶链反应(RT-PCR)方法测定CAT和GSH-Px mRNA的表达水平。结果 经USMF12 T照射12 h后,小鼠CATmRNA及GSH-Px mRNA的平均表达水平分别为1.154、0.881,高于对照组0.895、0.683;照射16 h组在照射后6 h,小鼠CAT和GSH-Px mRNA表达水平分别为(1.112±0.217)、(0.887±0.217),高于对照组(0.923±0.089)、(0.642±0.073)(P<0.05)。结论 USMF照射可使小鼠CAT和GSH-Px基因的表达发生变化。

     

    Abstract: Objective To study the effect of ultra strong static magnetic field(USMF) on gene expressions of catalase (CAT) and glutathione peroxidase(GSHPx) in mice.Methods One-week-aged mice were exposed to USMF at 12 Tesla (T) for 8,12 or 16 hours.Total liver RNAs was then extracted.Expression levels of CAT mRNA and GSH-Px-mRNA were mea sured with quantitative RTP-CR6 12 or 24 hours after the exposure.Results CAT-mRNA and GSH-Px-mRNA levels in 12hr-treated mice incresed to 1.154 and 0.881(optical absorption ratio) without significant increase compared to those of the controls.CAT-mRNA and GSH-Px-mRNA levels in 16hr-treated mice was upregulated (1.112 and 0.887,optical absorption ratio) 6 hours after the exposure.Conclusion Gene expressions of CAT and GSH-Px,the markers of antioxidant ability,are not impaired by 12TUSMF treatment in mice.
